N. sativa Oil Anti-Cancer Impact on MCF7 & AGS Cells

Irfan Çınar and his team aimed to explore the impact of N. sativa oil on various aspects of breast and gastric cancer cells, including cytotoxicity, apoptosis, invasion, metastasis, and heat shock proteins (HSPs).

Researchers performed an inclusive analysis of the cytotoxic and apoptotic effects induced by various concentrations (10-50-100-200 µg/mL) of N. sativa oil on MCF7 breast cancer and AGS gastric adenocarcinoma cell lines over 24, 48, and 72 hours using the MTT test. Additionally, the expression levels of Caspase-3, BCL2/Bax, MMP2-9, and HSP60-70 genes were assessed via RT-PCR in the treated cell lines.

About the MTT experiments, it was observed that N. sativa exhibited a time and dose-dependent inhibitory effect on MCF7 and AGS cancer cell proliferation. The vitality rates of treated MCF7 and AGS cells decreased to 77.04-67.50% at 24 hours, 65.28-39.14% at 48 hours, and 48.95-32.31% at 72 hours, with the doses of 100 and 200 µg/mL demonstrating the highest efficacy.

RT-PCR analysis revealed elevated caspase-3 levels and suppressed BCL2/Bax levels in both cell lines at higher concentrations of N. sativa. Additionally, exposure to N. sativa led to a notable decrease in MMP2-9 and HSP60-70 gene expression over time, particularly evident at a dosage of 200 µg/mL compared to the control group (P < 0.05).

The study concluded that N. sativa oil exhibits a dose-dependent impact on cytotoxicity and the expression of apoptotic, HSPs, and matrix metalloproteinases genes in both breast and gastric cancer.
